a chemical conjugate of the affinity-purified rabbit polyclonal antibody p75NTR (Cat. #AB-N01AP)
and the ribosome-inactivating protein, saporin

(molecular weight 210 kDa)

Conjugation of a p75NTR antibody to saporin has produced a cytotoxin that eliminates the CBF neurons, while sparing neighboring neurons that express GAD, calbindin and parvalbumin. This molecule eliminates cholinergic neurons in the basal forebrain of mice in the same manner that 192-IgG-SAP does in the rat.

    Thank you for your interest in our immunotoxin, mu p75-SAP, (Cat. #IT-16).  As you may know, this product has been on backorder for the past five months. The delay in producing mu p75-SAP was caused by the declining specificity of the antibody we were using as the targeting agent, a rat monoclonal to the mouse NGFr. After several failed attempts to regenerate this monoclonal, we decided to use a different antibody for the targeting agent. The new and more active version of mu p75-SAP utilizes an affinity-purified rabbit polyclonal to the mouse NGFr (Cat. #AB-N01AP). As you will see in the data sheet for the new version of this immunotoxin, the ED50 for the new product is in the picomolar range compared to an ED50 in the nanomolar range for the previous product (see “Previous Data Sheet”).

The new kit includes equal aliquots of saporin and the affinity-purified rabbit polyclonal antibody.  The kit and antibody will be available on April 20; the immunotoxin alone is available immediately.
