Conjugation of a p75^NTR antibody to saporin has produced a cytotoxin that eliminates the CBF neurons, while sparing neighboring neurons that express GAD, calbindin and parvalbumin. This molecule eliminates cholinergic neurons in the basal forebrain of mice in the same manner that 192-IgG-SAP does in the rat.

Elimination of Specific Cell Type

• Mouse p75^NTR-positive cells

To eliminate p75^NTR-expressing cells in rat, use 192-IgG-SAP. To eliminate p75^NTR-expressing cells in other mammals, use ME20.4-SAP.

Effective Tool to Study Behavior

Permanent and selective removal of cholinergic forebrain neurons makes an important animal model and can be used in conjunction with knockout models for the study of the following:

• Behavior (cholinergic forebrain function)
• Neuronal loss (e.g. Alzheimer’s disease)
• Plasticity of other systems in response to loss
• Replacement therapy
• Drug effects and dependence
